چکیده

With slices derived from two areas of guinea pig brain, cerebellum, and cerebrum plus brain stem, we have shown that specific, and different, receptors exist in these two brain areas for the control of cyclic adenosine 3’,5’-monophosphate (cyclic AMP) levels. The cerebellum contains a single type of receptor, shown to be a classical /3-adrenergic receptor. The remainder of the brain was shown to have at least two types of receptors mediating increases in cyclic AMP. One, specific for catecholamines, was shown to be a classical cr-adrenergic receptor. The other, specific for histamine, could be blocked by a number of antihistaminic agents. This report is the first to show that both CYand P-adrenergic stimulation can increase cyclic AMP levels.

Involvement of A pertussis Toxin Sensitive G-Protein in the Inhibition of Inwardly Rectifying K+ Currents by Platelet-Activating Factor in Guinea-Pig Atrial Cardiomyocytes

Platelet-activating factor (PAF) inhibits single inwardly rectifying K(+) channels in guinea-pig ventricular cells. There is currently little information as to the mechanism by which these channels are modulated. The effect of PAF on quasi steady-state inwardly rectifying K(+) currents (presumably of the I(K1) type) of auricular, atrial and ventricular cardiomyocytes from guinea-pig were studie...

The Role of Nitric Oxide and Prostaglandins in the Effect of Adenosine on Contractility, Heart Rate and Coronary Blood Flow in Guinea Pig Isolated Heart

It is a well-established fact that adenosine and its receptor subtypes (A 1 and A ) are involved in changes of contractility, heart rate and coronary blood flow (CBF) under different circumstances. This study was conducted to evaluate the role of nitric oxide and prostaglandins in development of these changes. For this purpose, Nitro-L-Arginine methyl ester (L-NAME), and indomethacin as inhibit...

Indirect and direct effects of the divalent cation ionophore A23187 on guinea pig and rat ventricular myocardium.

The contractile and biochemical effects of the divalent cation ionophore A23187 were studied in isolated perfused guinea pig and rat ventricles. A23187 increased both cyclic adenosine 3':5'monophosphate (cyclic AMP) and cyclic guanosine 3':5'-monophosphate (cyclic GMP) concentrations in guinea pig ventricles. Associated with these changes in nucleotide content were positive inotropic effects an...

Dissociations between changes in myocardial cyclic adenosine monophosphate and contractility.

The relationship between changes in the myocardial concentration of adenosine 3':5'-cyclic monophosphate (cyclic AMP) and cardiac contractility was studied in guinea pig and rat myocardium. When isolated perfused guinea pig heart were perfused with 10-5-M papaverine, a potent inhibitor of cyclic AMP phosphodiesterase activity, myocardial cyclic AMP concentration increased significantly from 1.7...

Is histamine potentiation of adenosine-stimulated cyclic AMP accumulation in guinea-pig cerebral cortical slices mediated by products of inositol phospholipid breakdown?

In several brain tissues the accumulation of cyclic AMP induced by directly acting stimulants, such as isoprenaline, adenosine or histamine (Hr) can be potentiated by histamine HI-receptor agonists [l-3]. The potentiation of adenosine-coupled cyclic AMP accumulation has proved useful in quantitative studies of functional H,-receptors in guinea-pig cerebral cortical slices [4, 51, but the recent...
